I say no. Not for Video recordings. Record a video an blow in the "mic" and you will see, its not in use.

I have 7 cases and only one has the hole - the carbon fiber one. I made the hole for my self by another one until I realized that the "mic" is not used for video. Even the Nillkin Cases dont have the hole. Maybe its used for noise canceling during calls.

The phone is recording stereo, the 3rd microphone is used to filter and improve audio while recording.

On calls only for noise canceling.
